Universal system for determining mutations in the promoter of the TERT gene on the example of the analysis of tumor lesions of the central nervous system

Abstract

Abstract. In this work, we compared real-time PCR and droplet digital PCR methods for the promoter region of TERT gene mutations detection using various systems of detecting probes. The detection limits of mutations for these methods were determined, based on which the advantages of the droplet digital PCR method for future studies of tumor lesions of the central nervous system were described. For the first time, the mutant allele fraction of changes in the promoter of TERT gene for cell lines of tumor lesions of the central nervous system was analyzed as 60% for glioblastoma Grade IV IDH1. To compare these methods, a versatile system based on recombinant plasmids was proposed, which proved to be better than the system based on genomic DNA.
